Pre-immune serum test of Rabbit

Materials: 1kg, male, New Zealand White

 

1.     After Purchase, adapt for 3 days

2.     Pick up the 300 ul blood from ear vein and centrifuge at 26,000Xg for 15 min

3.     Take supernatant(this is pre-immune serum)è Store at 4 degree for short term or -70 degree for long term storage

4.     Load human protein extracts (HeLa or HEK 293 etc..) and mouse protein extracts (NIH3T3 or MEF) into 11% SDS-PAGE

5.     After transfer your protein onto PVDF membrane, cut your membrane into N of test rabbit.

6.     Perform the Western blot : For primary antibody solution : 1/1500 dilution of preimmune serum

7.     When you expose the membrane into film, expand the exposure time.

 

    8. If you catch the band in the film of western with pre-immune serum, discard the Rabbit.
